Yield conditions for deformation of amorphous polymer glasses
Shear yielding of glassy polymers is usually described in terms of the
pressure-dependent Tresca or von Mises yield criteria. We test these criteria
against molecular dynamics simulations of deformation in amorphous polymer
glasses under triaxial loading conditions that are difficult to realize in
experiments. Difficulties and ambiguities in extending several standard
definitions of the yield point to triaxial loads are described. Two
definitions, the maximum and offset octahedral stresses, are then used to
evaluate the yield stress for a wide range of model parameters. In all cases,
the onset of shear is consistent with the pressure-modified von Mises
criterion, and the pressure coefficient is nearly independent of many
parameters. Under triaxial tensile loading, the mode of failure changes to

Growth, microstructure, and failure of crazes in glassy polymers
We report on an extensive study of craze formation in glassy polymers.
Molecular dynamics simulations of a coarse-grained bead-spring model were
employed to investigate the molecular level processes during craze nucleation,
widening, and breakdown for a wide range of temperature, polymer chain length
, entanglement length and strength of adhesive interactions between
polymer chains. Craze widening proceeds via a fibril-drawing process at
constant drawing stress. The extension ratio is determined by the entanglement
length, and the characteristic length of stretched chain segments in the
polymer craze is . In the craze, tension is mostly carried by the
covalent backbone bonds, and the force distribution develops an exponential
tail at large tensile forces. The failure mode of crazes changes from
disentanglement to scission for , and breakdown through scission
is governed by large stress fluctuations. The simulations also reveal
inconsistencies with previous theoretical models of craze widening that were
based on continuum level hydrodynamics
Intermittency in aging
The fluctuation-dissipation relation (FDR) is measured on the dielectric
properties of a gel (Laponite) and of a polymer glass (polycarbonate). For the
gel it is found that during the transition from a fluid-like to a solid-like
state the fluctuation dissipation theorem is strongly violated. The amplitude
and the persistence time of this violation are decreasing functions of
frequency. Around it may persist for several hours. A very similar
behavior is observed in polycarbonate after a quench below the glass transition
temperature. In both cases the origin of this violation is a highly
intermittent dynamics characterized by large fluctuations. The relevance of

ReferencesGold nanoparticles (Au NPs) and gold-based nanomaterials combine unique properties relevant for medicine, imaging, optics, sensing, catalysis, and energy conversion. While the Turkevich–Frens and Brust–Schiffrin methods remain the state-of-the-art colloidal syntheses of Au NPs, there is a need for more sustainable and tractable synthetic strategies leading to new model systems. In particular, stabilizers are almost systematically used in colloidal syntheses, but they can be detrimental for fundamental and applied studies. Here, a surfactant-free synthesis of size-controlled colloidal Au NPs stable for months is achieved by the simple reduction of HAuCl4 at room temperature in alkaline solutions of low-viscosity mono-alcohols such as ethanol or methanol and water, without the need for any other additives. Palladium (Pd) and bimetallic AuxPdy NPs, nanocomposites and multimetallic samples, are also obtained and are readily active (electro)catalysts. The multiple benefits over the state-of-the-art syntheses that this simple synthesis bears for fundamental and applied research are highlighted.Peer reviewe
The Mast Cell Degranulator Compound 48/80 Directly Activates Neurons
Background
Compound 48/80 is widely used in animal and tissue models as a “selective” mast cell activator. With this study we demonstrate that compound 48/80 also directly activates enteric neurons and visceral afferents.
Methodology/Principal Findings
We used in vivo recordings from extrinsic intestinal afferents together with Ca++ imaging from primary cultures of DRG and nodose neurons. Enteric neuronal activation was examined by Ca++ and voltage sensitive dye imaging in isolated gut preparations and primary cultures of enteric neurons. Intraluminal application of compound 48/80 evoked marked afferent firing which desensitized on subsequent administration. In egg albumen-sensitized animals, intraluminal antigen evoked a similar pattern of afferent activation which also desensitized on subsequent exposure to antigen. In cross-desensitization experiments prior administration of compound 48/80 failed to influence the mast cell mediated response. Application of 1 and 10 µg/ml compound 48/80 evoked spike discharge and Ca++ transients in enteric neurons. The same nerve activating effect was observed in primary cultures of DRG and nodose ganglion cells. Enteric neuron cultures were devoid of mast cells confirmed by negative staining for c-kit or toluidine blue. In addition, in cultured enteric neurons the excitatory action of compound 48/80 was preserved in the presence of histamine H1 and H2 antagonists. The mast cell stabilizer cromolyn attenuated compound 48/80 and nicotine evoked Ca++ transients in mast cell-free enteric neuron cultures.
Conclusions/Significance
The results showed direct excitatory action of compound 48/80 on enteric neurons and visceral afferents. Therefore, functional changes measured in tissue or animal models may involve a mast cell independent effect of compound 48/80 and cromolyn
Neuronal Conduction of Excitation without Action Potentials Based on Ceramide Production
International audienceBACKGROUND: Action potentials are the classic mechanism by which neurons convey a state of excitation throughout their length, leading, after synaptic transmission, to the activation of other neurons and consequently to network functioning. Using an in vitro integrated model, we found previously that peripheral networks in the autonomic nervous system can organise an unconventional regulatory reflex of the digestive tract motility without action potentials. METHODOLOGY/PRINCIPAL FINDINGS: In this report, we used combined neuropharmacological and biochemical approaches to elucidate some steps of the mechanism that conveys excitation along the nerves fibres without action potentials. This mechanism requires the production of ceramide in membrane lipid rafts, which triggers in the cytoplasm an increase in intracellular calcium concentration, followed by activation of a neuronal nitric oxide synthase leading to local production of nitric oxide, and then to guanosine cyclic monophosphate. This sequence of second messengers is activated in cascade from rafts to rafts to ensure conduction of the excitation along the nerve fibres. CONCLUSIONS/SIGNIFICANCE: Our results indicate that second messengers are involved in neuronal conduction of excitation without action potentials. This mechanism represents the first evidence-to our knowledge-that excitation is carried along nerves independently of electrical signals. This unexpected ceramide-based conduction of excitation without action potentials along the autonomic nerve fibres opens up new prospects in our understanding of neuronal functioning
Phase I dose escalation study of BI 836826 (CD37 antibody) in patients with relapsed or refractory B-cell non-Hodgkin lymphoma
BI 836826 is a chimeric immunoglobulin G1 antibody targeting CD37, a tetraspanin transmembrane protein predominantly expressed on normal and malignant B cells. This phase I, open-label study used a modified 3 + 3 design to evaluate the safety, maximum tolerated dose (MTD), pharmacokinetics, and preliminary activity of BI 836826 in patients with relapsed/refractory B cell non-Hodgkin lymphoma (NHL; NCT01403948). Eligible patients received up to three courses comprising an intravenous infusion (starting dose: 1 mg) once weekly for 4 weeks followed by an observation period of 27 (Course 1, 2) or 55 days (Course 3). Patients had to demonstrate clinical benefit before commencing treatment beyond course 2. Forty-eight patients were treated. In the dose escalation phase (1-200 mg) involving 37 Caucasian patients, the MTD was 100 mg. Dose-limiting toxicities occurred in four patients during the MTD evaluation period, and included stomatitis, febrile neutropenia, hypocalcemia, hypokalemia, and hypophosphatemia. The most common adverse events were neutropenia (57%), leukopenia (57%), and thrombocytopenia (41%), and were commonly of grade 3 or 4. Overall, 18 (38%) patients experienced infusion-related reactions, which were mostly grade 1 or 2. Preliminary evidence of anti-tumor activity was seen; three patients responded to treatment, including one complete remission in a Korean patient with diffuse large B cell lymphoma. BI 836826 plasma exposure increased more than proportionally with increasing doses. BI 836826 demonstrated preliminary activity; the most frequent adverse events were hematotoxicity and infusion-related reactions which were manageable after amending the infusion schedule. Although BI 856826 will not undergo further clinical development, these results confirm CD37 as a valid therapeutic target in B cell NHL

The variations in the heat of adsorption during the hydration, of homoionic montmorillonites are obtained by direct calorimetric measurements. The curves can show two important maxima ; the latter represent, in the course of successive swellings of the clay minerals, the initial hydration of the balance cation and of its solvation complement, when passing into the adsorbed phasis. The form, importance and position of these maxima depend on the nature of the cation and on the preliminary thermal treatment of the sample. They do not only inform about the necessary hydration energy but also about the transformations of the solid (swelling migration of the cation).Les variations de la chaleur d'adsorption au cours de l'hydratation des montmorillonites homoioniques sont obtenues par mesures directes calorimétriques. Les courbes peuvent présenter deux maxima importants qui traduisent au cours des gonflements successifs de l'argile l'hydratation première du cation compensateur et son complément de solvation quand il passe dans la phase adsorbée. La forme, l'importance et la position de ces maxima dépendent de la nature du cation et du traitement thermique préalable subi par l'échantillon.
